Hydroxysafflor yellow A promotes osteogenesis and bone development via epigenetically regulating β-catenin and prevents ovariectomy-induced bone loss

In clinical treatment, there is increasingly prevalent that traditional Chinese medicine treats common bone diseases including osteoporosis. Hydroxysafflor yellow A (HSYA), one of the essential compounds Safflower, has been used as therapy for thrombus, myocardial ischemia, and inflammation, but its effect on osteogenesis through epigenetic control ovariectomy-induced loss in vivo not explored. Therefore, study aimed to explore function mechanism HSYA formation development. We found could enhance cell viability promote hBMSCs vitro. Mechanistically, increase expression β-catenin leading accumulation nucleus activation downstream targets osteogenesis. Besides, RNA-seq quantitative RT-PCR western blot showed KDM7A was significantly increased by HSYA. The occupancy H3K27me2 promoter decreased HSYA, which be reversed silencing endogenous KDM7A. More importantly, promoted development chick embryos prevented ovariectomy (OVX)-induced SD rats. Taken together, our shown convincing evidence via epigenetically regulating prevent loss.
